The Centre of Excellence in New Target Discovery (CENTD), a partnership between FAPESP,
Glaxo-SmithKline (GSK) and the Butantan Institute, is offering a FAPESP Post-Doctoral
scholarship, available for immediate start.
The fellow will have the opportunity to work on the identification and characterization of new
molecular targets identified in cellular models using multiple approaches. In this process, cells
are subjected to various stimuli and then treated with potentially anti-inflammatory synthetic
peptides. Targets are identified by chemoproteomic strategies.
The fellow will focus primarily on validating the identified targets with the use of genomic
editing techniques such as CRISPR/Cas9 (gene knockout), RNAi gene silencing (knockdown)
and/or overexpression of genes of interest.
Candidates must have a PhD in Biochemistry, Molecular Biology or related areas, good
scientific output with publications as first author in the area of expertise that are compatible
with the position, motivation to solve complex biological problems in the field of molecular
target discovery, and the hability to work in an interdisciplinary environment. Excellent
organizational skills with experience in planning and developing experiments, a proactive
mindset, Independence and group work are essential. In addition, the candidate must have
good communication skills in English, for drafting scientific manuscripts and orally
presenting/discussing results at regular meetings with the participation of GSK.

The applicant will work with multidisciplinary teams for the identification of inflammatory events triggered by SARS-CoV-2, in different cell models, and the regulation of these events by the use of molecules derived from animal secretions, potentially antiviral drugs, monoclonal antibodies and mesenchymal stem cells. Requirements: Ph.D. in Biochemistry, Virology or related field, with at least 3 articles as the first author in international journals, motivation to solve complex biological problems in the field of cell biology/virology, motivation to work in an interdisciplinary environment, fluency in English with good writing ability. Project Title: Signaling pathways involved in the inflammatory response in 3D culture models of osteoarthritis
The Centre of Excellence for Research in Target Discovery, a new partnership among FAPESP, Glaxo-Smith Kline and Butantan Institute, has a postdoctoral position available in the Laboratory of Immunochemistry at Butantan Institute, Sao Paulo, Brazil.
The postdoctoral fellow will have the opportunity to work with a project on the identification/elucidation and validation of novel targets/pathways involved in osteoarthritis, using as tools toxins from poisonous animals.
As requirements, the candidates should have PhD in Immunology, at least three first author papers in international peer reviewed journals, motivation to solve complex biological problems in the field of target discovery and to productively working in an interdisciplinary environment and be fluent in English with good writing skills.
Good experience in 3-D cultures and in immunological techniques, including multi-color flow cytometry are essential skills required for the candidates. Experience in molecular biology and bioinformatics is desired.

Project
The inflammatory response involved in osteoarthritis will be studied in 3D culture models. This model allows to closely reproducing the microenvironment of the joints, once different cell types can be co-cultured, such as chondrocytes, osteocytes and fibroblasts. Immune cells can also be added to these models, allowing the study of their roles on the pathogenesis of osteoarthritis. Cultures will be stimulated by toxins isolated from different venomous animals, whose pro-inflammatory activities had been previously described, and the expression and production of molecules involved with inflammation will be evaluated.
After this initial in vitro screening, some toxins will be chosen for better elucidate the signaling pathways triggered in the different cell types present in the 3D culture. The study of signaling pathways will involve different assays, such as the use of specific inhibitors and the intracellular staining of signaling molecules, which will be analyzed by multicolor flow cytometry and confocal microscopy, including co-localization and tracking assays. These studies will enable the identification of novel signaling molecules involved with the inflammatory response, possibly novel targets for the development of anti-inflammatory drugs specific for the treatment of osteoarthritis.
